Caffeine Half-Life Explained, and How d9-Caffeine Changes It

"How long does caffeine last?" is really a question about half-life. Once you understand that one number, you can predict when your coffee will fade, why the same cup affects people so differently, and why d9-caffeine needs a different schedule.

What half-life means

A half-life is the time it takes for the amount of a substance in your blood to fall by half. After one half-life, half is left. After two, a quarter. After three, an eighth. Caffeine does not switch off; it fades on this curve.

Caffeine's half-life

In healthy adults, caffeine's half-life is commonly cited at around five hours, with a wide range between people. In one small 1986 study of six healthy men, it averaged about 4.1 hours. Here is what a five-hour half-life looks like for a 200 mg dose taken at 8 a.m.:

That leftover caffeine matters. In a well-known 2013 study, a large 400 mg dose of caffeine taken six hours before bedtime still cut total sleep by more than an hour.

Why it varies so much

  • Genetics. Variants in the gene for CYP1A2, the liver enzyme that clears most caffeine, make some people fast metabolizers and others slow. Our guide to fast and slow caffeine metabolizers goes deeper.
  • Smoking speeds caffeine clearance.
  • Pregnancy and some oral contraceptives slow it considerably.
  • Certain medications slow it by blocking CYP1A2.
  • Liver health affects it, since the liver does the work.

How d9-caffeine changes the timeline

d9-caffeine is caffeine with nine hydrogen atoms replaced by deuterium, placed exactly where CYP1A2 starts breaking caffeine down. Deuterium bonds break more slowly, so the molecule stays intact longer. In a 2022 double-blind study in 20 healthy adults, d9-caffeine was absorbed just as quickly as regular caffeine but produced four to five times the total exposure over time. Its developer describes it as lasting more than three times as long as ordinary caffeine.

Timing rules for d9-caffeine

  • Take it in the morning. A longer tail is useful by day and unhelpful at night.
  • Start with one capsule and see how your evening and sleep feel before adding more.
  • Do not redose in the afternoon the way you might with coffee. The first dose is still working.
  • If you are a slow metabolizer, pregnant, or on a medication that slows caffeine clearance, talk to your physician before using it.
